== Contraindications == Etifoxine is contraindicated in people with circulatory shock, severe liver impairment, severe kidney impairment, myasthenia gravis, galactosemia (due to lactose in the drug formulation), severe respiratory failure, and hypersensitivity (allergy) to etifoxine. The medication is not recommended in children or adolescents under the age of 18 and is not recommended during pregnancy and breastfeeding due to insufficient data. Caution is warranted with regard to combining etifoxine and other central depressants such as benzodiazepines, central analgesics, antipsychotics, sedative antihistamines, and alcohol.

== Cyclic Tetrapeptides == Cyclic tetrapeptides are a class of drugs that contain an α-epoxyketone group that has the potential to alkylate the HDAC active site. The HDAC active site, also known as histone deacetylase, are isozymes that modulate numerous regulatory signals and pathways within biological systems. They serve as targets for drug design. If the cyclic tetrapeptides were to alkylate the HDAC active site, they would deactivate the HDAC catalytic pocket. The tetra-peptide tuftsin (Thr–Lys–Pro–Arg), has been reported to affect a wide variety of biological responses in neutrophils and mononuclear phagocytes and also phagocytosis. It has also been reported that a tetra-peptide with the amino acid sequence, RGDS, that is from the cell-binding domain of the fibronectin molecule, is capable of blocking fibronectin from attaching to the cells. Based on that report, they were able to suggest that the RGDS tetra-peptide is capable of blocking RPE attachment to a variety of extracellular matrix component including; fibronectin, type I collagen, type II collagen, laminin, and lens capsule basement membrane. By utilizing time-lapse cinematography, it has been shown that the RGDS tetra-peptide inhibits the ability of cells to contract collagen.
== Research == Multiple studies have shown that LDN has promise as a treatment for chronic pain, some autoimmune disorders, and cancers. Studies on the efficacy of LDN for multiple sclerosis have been inconclusive. Clinical trials for its use as treatment of fibromyalgia were initiated in 2021. LDN is also being studied in long COVID. A 2018 therapeutic utilization review concluded that LDN may be an appropriate option for treatment of fibromyalgia and irritable bowel syndrome, but that "proper clinical trials are needed in order to establish evidence that could lead to correct indications, mode of administration, and other aspects necessary for effective clinical pharmacology of [low-dose naltrexone]." The UK's National Health Service echoed this sentiment in 2020. In a 2013 paper published in Arthritis & Rheumatology, authors Jarred Younger and Sean Mackey reported reduced circulating pro‑inflammatory cytokines and self‑reported pain in fibromyalgia patients using LDN. A 2018 review summarized mixed trial outcomes and emphasized methodological limitations.
